It is thus a protein substrate that donates only lysine side chains to the enzymatic reaction; this behavior was also seen with purified aB-crystallin, which, in the presence of guinea pig liver transgiutaminase, reacted only with dns-e-aca-QQIV but not with dansylcadaverine [J.B. Smith, P.T.V., and L.L., unpublished data]. The main purpose of the current work was to explore the usefulness of the approach of dissecting acceptor-donor relationships in the transglutaminase-mediated cross-linking of proteins for the rapid identification of relevant amino acid sequences. This is illustrated in the present report by the isolation of a prominent lysine-donating fragment in bovine lens. The labeling reaction with intrinsic transglutaminase was followed by centrifugation, which separated the high molecular weight polymers and cytoskeletal proteins from the crystallins 11 ; . F o r steers infused with lysine alone, the breakpoint occurred at 4 g infused lysine, whereas steers receiving 9 g d DL-methionine had a breakpoint at 8 g infused lysine. The total absorbable lysine requirement was estimated to be 44 depending upon methionine status. Rumen-protected amino acids fed to steers on both dietary regimens were effective in supplying methionine and lysine postruminally.

LYSINE R E Q PIGS and water were supplied ad libitum. Body weight and feed consumption were recorded biweekly. Composition of the basal diet for trials 1 through 4 is shown in table 1. L-lysine, as L-lysine HC1, was substituted isonitrogenously for L-glutamic acid to provide the dietary levels of lysine. On an isonitrogenous basis one unit of actual lysine replaced approximately two units of glutamic acid. Starch was added to bring the sum of lysine plus starch equal to the quantity of glutamic acid replaced. For substitution purposes lysine, glutamic acid, and starch were considered essentially isocaloric. Sodium bicarbonate was added on a molar basis with glutamic acid to neutralize the second acidic group of glutamic acid. Since sodium bicarbonate and the HC1 of L-lysine HC1 were assumed to contain little energy, sand was added to bring the sum of sodium bicarbonate, sand and HCI to a constant amount in all diets. The diets assayed 13.3% crude protein and 3, 501 kcal kg of metabolizable energy. ABSTRACT Experimentswere conducted withyoung pigs to evaluate the effects of excess lysine when added to diets marginal in arginine. The basal diet was formulated to con tain 1.15% lysine and 0.53% arginine. Lysine levels above this were achieved by incremental additions of L-lysine HCI experiment 1 ; or L-lysine acetate experiment 2 ; . Neither weight gain nor gain-feed ratio was lowered in pigs fed twice the recommended level of lysine i.e., 2.30% ; . Lysine levels three or four times the basal level, however, lowered both weight gain and feed intake with no commensurate reduc tion in feed efficiency. Therefore, lysine appeared to reduce growth via amino acid imbalance rather than antagonism. In pigs equally fed isonitrogenous diets, urea N excretion in the urine was lower in those fed 3.45% supplemental lysine 4.6% total lysine ; than in those fed the basal diet. This was offset by a dramatic elevation in urinary excretion of basic amino acids. Urinary excretion of lysine accounted for 26.2% of the lysine consumed by pigs fed 3.45% sup plemental rysine. Free lysine levels as well as a-aminoadipic acid levels were markedly higher in plasma, liver, kidney and muscle of pigs fed excess rysine than in those fed the basal diet.
