The use of genetic resistance by breeders in breeding chickens less susceptible to disease causing pathogens. We have initiated a random-primer PCR-based genome-wide scan of lines divergently selected for immune response to sheep red blood cells. Using DNA templates from high and low-line birds in the 27th generation of selection for immune response, we have screened a total of 12 primers for their informativeness in the selected lines. While ten of the 12 primers amplified fragments, only 8 or 67% amplified polymorphic fragments. The size of the polymorphic fragments has ranged from 600 base pairs to 3500 base pairs. Though it is early to establish any relationship with immune response, which will be done in future work, the lines appear to be highly heterogeneous as indicated by the RAPD fragments and may pose a problem in QTL analysis. This suggests an approach that has been widely used to identify genes for complex traits in plants can be efficiently used in screening the appropriate animal populations.
