The discrepancies may be caused by variation in susceptibility of isolates in some centres or by methodological problems. The latter is most likely. Interpreting indistinct endpoints by double zones with co-amoxiclav may have influenced the Etest results for H. influenzae, 8 recording only 67% susceptibility versus 98% with the broth dilution method, the latter fitting well with the -lactam resistance among H. influenzae strains in The Netherlands 5% ; . We concluded, therefore, that the Etest results for co-amoxiclav were unreliable.

Firstly, negatively-charged modification can hinder proteolytic digestion by trypsin and secondly, the sequence near the phosphorylation site is usually very hydrophilic, leading to a selective loss of phosphopeptides during the standard methods used for sample preparation. Thirdly, mass spectrometers have a finite dynamic range, peptides of very low abundance may not be picked up for sequencing in and stadol.

In complex I from this mutant iron-sulfur cluster N2 was not detectable, and all other EPR-detectable iron-sulfur clusters were unchanged Fig. 2 ; . Mutation D458A also resulted in about 70% loss of activity, and EPR spectra of complex I from D458A were normal. With IC50 value increases of 30-fold for DQA and 10-fold for rotenone, this mutant exhibited by far the most pronounced inhibitor resistance Table I; Fig. 3 ; .
