In April 2006, Birmingham OwnHealth was launched, to enrol 2, 000 patients in telephone-based care. Objectives: The goal is to better understand the genomic regulatory mechanisms acting on xenobiotic transporters. It is expected that a comprehensive examination of regulatory mechanisms of representative transporters, such as the multidrug resistance-associated protein-3 MRP-3 ABCC3 ; , will have broader implications and provide a frame of reference for a more general viewpoint of these molecules. MRP3, is an ATP-binding cassette ABC ; protein transporting organic anions in multiple tissues, notably involved in the cellular export of glucuronide conjugates and clearing of toxic bile acids in the liver. Approaches: The laboratory will take advantage of comparative genomic and experimental cellular approaches to identify potentially new response elements that may be present there. Experimental verification of functions of coding or noncoding, regulatory genomic sequences often depends on cell culture approaches The laboratory has established the first cartilaginous fish cell line SAE ; , a mesenchymal stem cell line derived from the embryo of an elasmobranch, the spiny dogfish shark Squalus acanthias. Development of the cell line was dependent on the Cell Culture and Animal Services Cores. Characterization of the line was aided by the Imaging Core. Elasmobranchs first appeared over 400 million years ago, and existing species provide useful models for comparative vertebrate cell biology, physiology and genomics. The approach is to use expressed-sequence tags ESTs ; to define mRNAs derived from the SAE cell line for identification of conserved regulatory regions in the noncoding portions of the mRNAs and to clone the nontranslated flanking regions of xenobiotic transporter genes to identify regulatory regions in these areas. Gene sequence will be determined by BAC identification using gene-specific primers deduced from ESTS, and cloning in collaboration with Dr. A. Planchart at MDIBL. Similar approaches will be applied to skate embryo cell lines analogous to the SAE cell line. The skate represents the most primitive tractable model of vertebrate development, allowing developmental studies. The molecular biological approaches described require the extensive use of the Instrumentation Core. Accomplishments: A survey of 5000 expressed sequence tags ESTs ; from the SAE cell line identified it as derived from mesenchymal stem cells. These ESTs also revealed genomic regions in common with holocephali, an order of even more primitive cartilaginous fish that diverged from elasmobranchs 500 million years ago. Using primers identifying xenobiotic transporters previously cloned from skate liver, MRP3 cDNA was isolated from SAE mRNA and the sequence has been nearly completed, including a 3' UTR of about 300 bp and a coding region of 1498 amino acids. The protein contains two ATPbinding domains and 13 transmembrane domains TMDs ; . As expected for MRP3, there is no PZD domain. Based on analogous structures from other species, we estimate that a region coding for approximately 30 amino acids and a 5' UTR of approximately 200 bp remain to be sequenced. The protein is 65% identical in amino acid sequence with human MRP3, with the most identity occurring in the TMDs. Among these, TMD 6 showed the least identity with human. Among the other regions, least identity was seen in the intracellular region between TMD 8 and 9. Also cloned from a skate embryo cell line was the organic solute transporter subunit beta OSTb ; , a G-protein-like molecule. Histochemistry with specific antibody provided in a collaboration with Dr. Ballatori showed that OSTb was expressed in all cells of the culture. OST beta also was found in a wide variety of skate tissues. Expression of the other subunit of this transporter, OST alpha, was limited to the liver, heart, intestine and gill. PROBLEM UNDER STUDY: Older adults are the fastest growing segment of the US population, causing many to question the possible safety impact of the growing number of older drivers on our roadways. In fatal crashes, the highest crash involvement rates are seen among the youngest and oldest drivers. The public health burden of these fatal crashes can be thought of as the sum of 1 ; the deaths and injuries among the drivers and 2 ; the deaths and injuries among all others involved in the crashes. OBJECTIVES: To assess the risk of death or non-fatal injury posed to other road users by older drivers versus drivers of different ages. METHOD OR APPROACH: Two-vehicle police-reported crashes for 1992-1994 were used to quantify the risk of death and injury that drivers of different ages impose on other road users. Crash-related deaths and injuries were separated into two categories, those occurring among the drivers themselves and those among others. Age of driver was grouped into six categories: 16-19 years, 20-34 years, 35-59 years referent ; , 60-74 years, 75-84 years, and 85 years and older. Drivers of all ages were involved in crashes that resulted in death or non-fatal injury to others. We calculated "excess risk" as the number of deaths or injuries injuries were estimated by transfers from the crash scene to emergency department facilities ; to others above those of drivers aged 35-59 years the referent group.

The arrival of a very welcome intruder at the Savoy during the Launch of a Million Prostate Miles was quickly spotted by our Royal Patron who had just made the first donation to this initiative. Prostaglandins are formed from the cyclization and peroxidation of arachidonic acid and have been shown to increase human sperm motility. The conversion of arachidonic acid into prostaglandins is catalyzed by cyclooxygenase COX ; . The effect of inhibition of prostaglandin synthesis on sperm mobility was tested with the addition of the COX inhibitors indomethacin, diclofenac sodium, aspirin, and tolmetin. Pooled turkey semen was diluted to 1 x 109 sperm mL with prewarmed motility buffer 50mM TES 128mM NaCl 2mM CaCl2 , pH 7.4; MB ; or MB containing COX inhibitors such that the final concentration was 0 to 10mM except for aspirin where 15mM was the highest dose ; . Diluted semen 0.06 mL ; was overlayered onto prewarmed 4% AccudenzTM solution 0.6 mL ; in 1.5 mL polystyrene cuvettes and incubated at 41 C for 5 min after which the absorbance at 550nm was recorded. Ten replicates were done for each drug. All compounds tested caused a decline in sperm mobility. Indomethacin and diclofenac showed increased inhibition of mobility 15 to 100% and 4 to 100%, respectively ; as the dose level increased. However, at 100% inhibition, sperm cells were no longer intact. For tolmetin and aspirin, there was a 27% and 22% decline in sperm mobility, respectively, at the lowest dose level 0.00lmM for tolmetin and 0.01mM for aspirin ; , but no further significant decline was seen until the 10mM dose for tolmetin 74% ; and 15mM dose for aspirin 42% ; . Addition of specific inhibitors of COX-1 resveratrol ; and COX-2 NS-398 ; at 0.1mM resulted in a 32% and 28% reduction in mobility, respectively. In conclusion, known COX inhibitors reduce sperm mobility in the turkey. The role of prostaglandins in turkey sperm mobility remains to be determined.
